The blotting

Transfer solution 8 is taken up by the stack of paper
towels 3. The DNA is transferred by the flow that goes through wick 6,
gel 5 and nylon filter (or "blotting membrane") 4. DNA molecules cannot pass that filter and are
bound to it. Solution 8 has a very high pH and will denature the DNA.
So, the DNA pattern of the gel is copied into a pattern of single-stranded DNAs on the
filter. This filter, now called a "blot",
can be used in a subsequent hybridisation experiment.. |
