A Cloning Course recombinant plasmid

A typical example of such a recombinant plasmid.

The Lambda DNA fragment has been cloned into E.coli, by using plasmid pGT4 as a vector.
The Lambda DNA fragment is inserted in the pGT4 plasmid.

We can obtain high amounts of the DNA fragment, just by growing a large culture of the E.coli clone and purify its plasmid DNA...

How do we get clones harbouring a recombinant plasmid?
